DELAYED HYPERSENSITIVITY III. SP~.cmlc DESENSITIZATION OF GUINEA PIGS SENSITIZED TO PROTEIN

چکیده
A general method for experimental induction in guinea pigs and man of the delayed hypersensitive state directed against protein antigens has been decribed previously (1). Sensitization follows a single intradermal injection of a small amount of antigen in the form of a washed immune precipitate. Provided the specific precipitation is carried out in the region of antibody excess, active antibody production is suppressed and no circulating antibody can be detected for 2 to 3 weeks after the delayed type of skin reactivity has appeared. If the sensitizing injection is suspended in adjuvant containing killed acid-fast bacteria, the delayed skin reactions to a given amount of antigen are larger than those seen when mycobacteria are excluded. The present studies were undertaken to determine the effects of a single injection of specific antigen on the subsequent skin reactivity of guinea pigs sensitized by this method. In this paper we have used the term desensitization, as have others in the past, to indicate loss of skin reactivity to a specific antigen. In the experiments to be reported it has been shown that guinea pigs sensitized to protein antigens can be completely and specifically desensitized by a single injection containing a sufficient amount of the corresponding antigen. In the following paper, the systemic effects of specific challenge are described (2).

Studies on Delayed Hypersensitivity
Experiments carried out with several well defined antigenic systems (hapten conjugates of poly-L-lysine and guinea pig serum albumin) in guinea pigs demonstrated that: 1. Arthus reactions also manifest carrier specificity, although to a smaller extent than do delayed hypersensitivity reactions. 2. Desensitization by injection of minute doses of antigen results in moderate specific desensitizati...
متن کاملPURIFICATION AND CHARACTERIZATION OF PROTEIN ANTIGENS ISOLATED FROM MYCOBACTERIUM TUBERCULOSIS (H37Rv STRA IN) AND THEIR EFFECTS ON CELL-MEDIATED IMMUNE RESPONSES IN GUINEA PIGS
Mycobacterium tuberculosis (H37Rv strain) was used in this study. The bacterial cells were disintegrated by sonication. The separation and characterization of the soluble molecules were attempted by various techniques including gel filtration, ion exchange chromatographies and polyacrylamide gel electrophoresis, using SDS and 2ME. Eight protein molecules with molecular weights ranging from...
متن کاملTHE EFFECT OF ASCORBIC ACID ON BLOOD HISTAMINE LEVEL AND DELAYED-TYPE HYPERSENSITIVITY IN GUINEA PIGS
The effect of ascorbic acid on blood histamine level and delayed-type hypersensitivity was studied in thirty male guinea pigs. Animals were sensitized to B.C.G., trichophytin and mallein during the first four weeks. The diet was ad libitum during this period. After this "sensitization period" animals w er e tested intradermally with tuberculin, trichophytin and mallein. The number and diam...

Objective(s): The effect of the extract of Crocus sativus and its constituent, safranal on inflammatory markers in sensitized guinea pigs was examined. Materials and Methods: Ovalbumin (OA) sensitized guinea pigs were given drinking water alone (group S), or drinking water containing three concentrations of safranal, three concentrations of extract and one concentration of dexamethasone, (n...

Objective: Pharmacological effects of carvacrol such as its anti-inflammatory activities have been shows. In this study the effects of carvacrol on serum levels of total protein (TP), phospholipase A2 (PLA2) and histamine in sensitized guinea pigs was evaluated. Materials and Methods: Sensitized guinea pigs were given drinking water alone (group S), drinking water containing three concentration...
